Réalisé par Aaron Horvath et Michael Jelenic (collaborateurs sur Teen Titans Go!, Teen Titans Go ! Le film) à partir d'un scénario de Matthew Fogel (Le film Lego 2, Les Minions 2: Il était une fois Gru), le film met en vedette Chris Pratt dans le rôle de Mario, Anya Taylor-Joy dans celui de la Princesse Peach, Charlie Day dans celui de Luigi. Jack Black jouera Bowser, Keegan-Michael Key interprétera le rôle de Toad, Seth Rogen celui de Donkey Kong, Fred Armisen jouera Cranky Kong, Kevin Michael Richardson sera Kamek et Sebastian Maniscalco interprétera Spike.
